Course Description
本課程擬介紹基礎核酸操作與蛋白操作等技術之學理給初入生物醫學研究領域的研究生,冀望研究生藉由瞭解技術背後的學理之後有能力自行解決實驗操作時所出現的問題,並得以進一步設計更適當的實驗操作方法。

Week 1 1. Biochemistry of restriction enzymes
Week 2 2. Biochemistry of DNA modifying enzymes
Week 3 3. Cloning vectors: plasmids, phagemids, cosmids, PAC, BAC, YAC
Week 4 4. Competent cells and transformation
Week 5 5. DNA sequencing
Week 6 6. Southern blotting and Northern blotting analysis
Week 7 7. Semi-quantitative RT-PCR and quantitative real-time RT-PCR
Week 8 8. Determination of protein concentration
Week 9 9. Cell lysate preparation: whole cells or fractionation
Week 10 10. Electrophoresis for DNA and proteins
Week 11 11. Western blotting analysis
