Course Description 隨著基因組解序技術的飛速進步與成本下降,基因體學已從單一基因的研究演變為現代生物學的核心工具,尤其在植物科學領域更顯關鍵 。相較於人類與動物模式生物擁有完善的基因體資源,植物物種繁多且基因體複雜,研究往往需從零開始,對特定物種進行序列解碼、組裝與註解,才能建立基礎研究工具 。本課程旨在帶領碩士班學生深入掌握從 DNA 定序、大數據序列分析、基因表現偵測,到差異表現基因群解析的完整流程,並探索後基因體時代(Post-genomic era)的研究趨勢 。

課程內容涵蓋四大核心領域:
(1) 基因體結構與定序策略:除了傳統桑格定序與次世代定序(NGS),課程將深入介紹第三代定序技術(如 PacBio SMRT 與 Nanopore)及光學圖譜技術,並探討 Telomere-to-Telomere (T2T) 的完整組裝策略 。學生將學習構建染色體圖譜、物理圖譜與遺傳圖譜,並應用多種 DNA 分子標誌(如 SNP, SSR)進行關聯性分析(GWAS)與分子輔助育種(MAS) 。
(2) 功能基因體學(Functional Genomics):課程涵蓋 mRNA 表現圖譜分析技術,包括微陣列(Microarray)與 RNA-Seq 的數據探勘(如 PCA 分析、GO enrichment) 。此外,將介紹高靈敏度的核酸定量技術,如即時定量 PCR (qPCR) 與數位 PCR (ddPCR) 的原理與應用 。
(3) 基因功能解析與編輯技術:探討反向遺傳學策略,包括 RNA 干擾(RNAi)、VIGS 等基因靜默技術 。重點單元將聚焦於 CRISPR/Cas9 基因編輯系統,從細菌免疫機制的原理、gRNA 設計、脫靶效應分析,到精準育種的實際應用案例,進行全方位的解析 。
(4) 實驗設計與實作思維:課程設有「學生主導之實驗設計工作坊」,引導學生針對特定植物基因,規劃包含基因靜默、過量表現或基因編輯的完整實驗藍圖,培養獨立研究與解決問題的能力 。

透過本課程,學生不僅能熟悉最前沿的基因體學技術與生物資訊資料庫資源 ,更將學會如何將這些工具轉化為解決植物科學問題的策略,為未來的學術研究或生技產業應用奠定堅實基礎。
